Characterization of global cfDNA fragmentomics in Melanoma Patients

MPM and SMPM patients were more often male and SMPM patients were older and presented with thicker melanomas than SPM patients.Mean follow-up was 8.4 (95%CI 8.3-8.4) years and mean time to second melanoma was 53.1 (95%CI 51.5-54.6)months.MSS at 20 years was 87.1% (95%CI 86.8-87.4),84.1% (95%CI 81.4-86.8),and 84.5% (95%CI 81.8-87.2) for SPM, MPM, and SMPM patients respectively. After adjustment in multivariable analyses, MPM was a risk factor for melanoma related death (HR: 1.32 (95%CI 1.16-1.49)),while SMPM was not (HR: 1.11 (95%CI 0.96-1.29)). Melanoma-specific furvival for SPM, MPM and SMPM patients in Denmark and Sweden 2000-2023Conclusions: This study found a significantly lower MSS in patients with MPM compared to patients with SPM, but no statistical difference in MSS between patients with SMPM and SPM.These findings warrant consideration of surveillance of MPM patients, regardless of tumor stage.
