“Isolation and Characterization of Steroidal Alkaloids from Sarcococca Saligna , Their Antinociceptive, Anti‐Inflammatory, and Gastroprotective Effect in Animal's Model”

Abstract In the current research, two steroidal alkaloids (holaphylline (HP) and sarcodine (SD)) were isolated from the methanol extract of Sarcococca saligna and evaluated for analgesic, anti‐inflammatory, and gastro‐protective activities in animal's model at dose of 5 mg/kg b.wt. Intraperitoneal administration of HP produced more significant (***P < 0.001) inhibition in writhing response while SD revealed slightly less than significant (**P < 0.01). In hot plate method, both compounds prolonged the paw licking almost same (***P < 0.001) as compared to control group. Furthermore, HP produced significant reduction in hind paw edema (**P < 0.01), while SD produced slight significant (*P < 0.05) reduction after 4 h of administration. Stomach histology and biochemical features showed that SD has maximum gastric lesions inhibition effect (***P < 0.001), while HP has significant effect (**P < 0.01) in aspirin induced gastric ulcer. Moreover, SD caused a significant decrease in gastric juice pH, while HP slightly decreased the gastric juice pH. SD showed no significant lesion score while HP showed slight lesion score (P < 0.05). Molecular docking indicates that these molecules may produce anti‐inflammatory effects by targeting the allosteric sites of COX‐1/COX‐2 enzymes. These findings signify strong analgesic and anti‐inflammatory effects of both compounds with beneficial effect against gastric ulcers.
