EVALUATION OF THE EFFICACY OF LEU-ILE-LYS TRIPEPTIDE IN THE CONTEXT OF CHRONIC ACETATE-INDUCED GASTROPATHY
Introduction. Peptic ulcer is a common multifactorial disease that significantly reduces the quality of life of a large number of patients. An obligatory stage in the development of new methods of pharmacological correction of this pathology is their experimental testing on models of acute and chronic gastropathy. Previously, studies were conducted on the basis of the Altai State Medical University, which demonstrated the Leu-Ile-Lys antiulcer effect of the tripeptide on models of acute gastropathy: indomethacin, ethanol and predniolone. For a complete preclinical assessment of the test substance, it is necessary to test it under conditions of modeling chronic gastric ulceration modeling. Objective. Study of the effect of Leu-Ile-Lys tripeptide on the course of experimental chronic "acetate" gastropathy. Materials and methods. The experiments were carried out on stock Wistar rats, divided into a disease control group, a treatment group that received Leu-Ile-Lys tripeptide at a dose of 11.5 mg / kg and a comparison group administered omeprazole at a dose of 37 mg/kg. In all groups, chronic acetate gastropathy was simulated, followed by intragastric administration of the studied substances for 5 days. At the end of the experiment, the stomachs of the experimental animals were extracted, revealing macro- and microscopic signs of the development of the pathological process. Results. The size of ulcerative defects compared to the same indicator of the disease control group was significantly decreased by 2.5 times in rats treated with the Leu-Ile-Lys tripeptide, while against the background of omeprazole administration it decreased by 1.7 times. According to the results of a pathohistological study in the Leu-Ile-Lys tripeptide treatment group, a decrease in inflammatory infiltration was recorded in the area of ulcerative defects, an increase in the number of collagen fibers and an increase in angiogenesis processes relative to the disease control groups and the administration of omeprazole. Conclusion. Thus, during the experimental testing of the Leu-Ile-Lys tripeptide against the background of chronic "acetate" gastropathy, the presence of an antiulcer effect of the substance under study was revealed, which is characterized by a decrease in the size of ulcerative defects, as well as activation of regeneration processes.
