"STRATOLAMP: LABEL-FREE, MULTIPLEX DIGITAL LOOP-MEDIATED ISOTHERMAL AMPLIFICATION BASED ON VISUAL STRATIFICATION OF PRECIPITATE"
Multiplex, digital nucleic acid detections have important biomedical applications such as pathogen detection, genetic analysis, and environmental monitoring. However, the multiplexity of existing methods predominantly rely on fluorescent dyes or probes, making the detection complicated and costly.Strategies for multiplex nucleic acid detection in a label-free manner are yet to be achieved.Leveraging the precipitate byproducts generated in loopmediated isothermal amplification (LAMP), we and other groups previously reported label-free, droplet digital LAMP technologies by analyzing the precipitate byproduct and achieved single target detection. 1, 2To expand its capability and achieve digital detection of multiplex targets, here we report the technology of StratoLAMP, based on the visual stratification of precipitate. 3
