MONITORING OF DRUG-IMFACTED CARDIOMYOCYTES CONTRACTILITY USING PI MICROCANTILEVER STRUCTURES WITH NANO-SILICON STRAIN SENSOR

Quantitative evaluation of cardiac contractility changes due to drugs is vital for early drug development.Cantileverbased biosensors, especially those integrated with strain sensors, offer real-time monitoring and high-throughput screening. However, existing cantilever biosensors, typically millimeter-sized, face challenges in sensitivity when miniaturized further.Additionally, SU-8, previously used in cantilever fabrication, may deform over time in solution, affecting reliability. Here, we introduce a novel method: polyimide (PI, KMSF-1000) cantilevers integrated with nano-silicon strain sensors for long-term in vitro cardiac toxicity assessment.Low water absorption and residual stress of PI enhance cantilever stability, while nano-silicon sensors improve accuracy. Our structure achieves a remarkable force detection limit of 1.3 nN m -2 .Tests with heart drugs verapamil and isoproterenol demonstrate the potential of the proposed structure for drug toxicity screening.

Abstract
