A CLEAN-UP-BASED MICROFLUIDIC SAMPLE PREPARATION WORKFLOW FOR LOW-INPUT PROTEOMICS
Mapping the proteome of low-input samples and single cells is crucial to elucidate mechanisms in disease states such as cancer. However, almost all previous efforts to miniaturize existing sample preparation protocols for low-input protein samples[1] are based on one pot in-solution methods, where lysis reagents need to be compatible with liquid chromatography-mass spectrometry (LC-MS).In contrast, clean-up methods enable washing steps and as such are compatible with stronger detergents, which provide more efficient cell lysis and protein solubilization [2].Here, we present for the first time different steps taken towards an integrated microfluidic approach for clean-up-based proteomic sample preparation for low-input samples.
