P-07.02 Pembrolizumab associated to chemotherapy in solid tumor:a retrospective study about 11 cases

Background
Chemotherapy has been the basis of advanced cancer treatment for many times. In the last decade, immunotherapy have revolutionized cancer treatment, becoming the frontline therapy for many cancers. In some tumors, such as melanoma, NSCLC and others, immunotherapy has largely replaced chemotherapy owing to its clinical benefits. Pembrolizumab, is a monoclonal antibody that belongs to a class of drugs blocking the PD-1/PD-L1 signaling pathway to enhance the anti-tumor immune response. It targets the PD-1 protein on the surface of immune T cells, essentially ‘taking the brakes off’ the immune system, to help eradicate tumors.
Materials and Methods
This is a retrospective study based on the medical records of 11 patients treated in the medical oncology department of Didouche Mourad Hospital, Constantine, Algeria. The patients received pembrolizumab during a period of 6 months; From July 2024 to December 2024.
Results
We enrolled 11patients.6 women for 5 men. The average age was 49 years (28–70 years). 45,45%(n=05) of patients had comorbidities. The most common cancer type was melanoma (n=08)72,72%, followed by the NSCLC in18,18% (n=02) and squamous cell carcinoma of the cervix in 9,09%(n=01). The most common metastatic site was liver found in 62,5% (n=05) of cases followed by the brain 37,5%(n=03) and the lung 25% (n=02).81,81%(n=08) of patients had received Pembrolizumab in first line, 25%(n=02) was associated to chemotherapy. 27,27%(n=03) of patients had received adjuvant Pembrolizumab in stage III resectable melanoma. Side effects occured in 18,18% (n=02) of patients, however 81,81% (n=09) of the patients tolerated the treatment very well. The first evaluation according to irRecist 1.1 was a partial response in 37,5% (n=03) of cases, stable disease in 25% (n=02) of cases, tumor progression in 24% (n=06) of cases. Currently, 72,72%(n=8) are undergoing treatment. Death occurred in 9.09% of patients (n=01). Our results showed that Pembrolizumab had an acceptable side-effect profile and antitumor activity in numerous malignancies.
Conclusions
Pembrolizumab has proven to be a revolutionary immunotherapy drug that has saved thousands of lives worldwide. Due to its potential for treating various types of cancer. Multiple clinical trials are underway to test the efficacy and success of pembrolizumab in different indications, either as a single agent or in combination with other therapies for different types of cancers.
Reference
Hamid O, Puzanov I, Dummer R, Schachter J, Daud A, Schadendorf D, Blank C, Cranmer LD, Robert C, Pavlick AC, Gonzalez R, Hodi FS, Ascierto PA, Salama AKS, Margolin KA, Gangadhar TC, Wei Z, Ebbinghaus S, Ibrahim N, Ribas A.
