P-08.03 Eliminating operator errors in flow analytics: dried reagents and fully automated flow gating provide fast and robust CAR T cell analytics

Background
In the realm of flow cytometry, particularly in the rapidly evolving field of cell and gene therapy (CGT) analytics, the demand for high-throughput data analysis and reproducibility is paramount. These requirements are driven by the necessity to ensure consistent results across diverse experimental setups and laboratories. Achieving high levels of standardization and reproducibility is crucial for advancing the capabilities of CGT analytics and setting new benchmarks in the field.
Materials and Methods
We introduce cutting-edge tools for cell analysis workflows, encompassing everything from staining procedures to automated flow cytometry analytics. Central to our approach is the use of comprehensive workflows, standardized acquisition protocols, and standardized dried antibody cocktails. These innovations include: Dried antibody cocktails: Minimizing variability, and drastically speeding up the assay protocol. Advanced Algorithms: Automating gating and population identification, significantly reducing manual intervention and human error. Pre-defined Experimental Settings: Allowing a standardized and quick machine setup. Programmed Layouts for Plots and Statistics: Providing ready-to-approve summaries for reports, which can be directly exported. User-friendly Interfaces: Enhancing convenience with comprehensive documentation features and robust audit-trail tracking.
Results
The implementation of these tools demonstrated a significant decrease in hands-on time and reduction of manual steps required, accelerating the staining procedure, acquisition, and analysis. The ready-to-use format of the dried reagents sped up the assay protocol, while the automatic gating suggestions required little to no manual adjustments, decreasing variability in the analysis.
Conclusions
Overall, the provision of results via a ‘push of a button’ enhanced the reliability and reproducibility of flow cytometry experiments and minimized operator variability. This mitigates risks of human errors and ensures comparability between different laboratory sites. By adopting these automated solutions, laboratories can achieve unparalleled efficiency and accuracy in flow cytometry analysis, advancing the capabilities of CGT analytics. E. Oldewurtel: A.
