INTEGRATING SCATAC-SEQ WITH IMAGING MODALITIES VIA MAGNETIC-BASED DETERMINISTIC BARCODING
Single cell approaches are essential for unveiling cell heterogeneity and biological mechanisms [1].Microwells represent a standard tool for imaging, and they can facilitate cell isolation prior to single cell assay for transposase-accessible chromatin with sequencing (scATAC-seq).To directly correlate chromatin accessibility profiles with cell or organelle morphology, both datasets need to be linked using barcodes, sequences that can spatially associate the scATAC-seq profile to the corresponding image.Current methods for barcoding like droplet printing are laborious and time consuming[2], while previous microfluidic-based deterministic barcoding strategies have focused on tissue sections [3,4], and are not readily adaptable to microwells due to the heightened risk of cross-talk.
