Real-time precise optical control of ATP uncaging in zebrafish (Conference Presentation)
To precisely control chemical processes in biological samples in a chemical-specific and site-specific manner, we have developed real-time optical control (RPOC) technology. This technology has demonstrated the capability to precisely manipulate the functions of individual organelles and activate photosensitive drugs within subcellular compartments. RPOC also allows for the concurrent creation of various optical treatment conditions within the same field of view (FOV) and enables comprehensive monitoring of both short-term and long-term cellular responses. However, previous RPOC studies have only been conducted on single cells. Here, for the first time, we apply RPOC in live zebrafish embryos to achieve precise uncaging of ATP molecules at single-cell resolution. The simultaneous opto-control and imaging capabilities of RPOC also allow us to photouncage ATP and monitor the generation and propagation of calcium waves triggered by the localized ATP release. This study demonstrates the potential application of RPOC for precise opto-control in live animals.
