Multispectral pulse-sampling fluorescence lifetime imager for high-speed large-scale tissue imaging

We developed a multispectral laser-scanning pulse sampling fluorescence lifetime imaging (PS-FLIm) imager for rapid, large-scale acquisition of time-resolved fluorescence data. The system, comprising a pulse laser, Galvo mirror, translation stage, and multiple optical components, achieved relatively high spatial resolution over a large area with fast scanning. The PS-FLIm imager was characterized using beam profiling and the USAF target card, and then used to image frozen sectioned pig coronary arteries. Autofluorescence lifetime (AFL) images were reconstructed from four spectral bands (390 nm, 470 nm, 540 nm, 630 nm). PS-FLIm imager can serve as a rapid-scanning tool for ex-vivo specimens. With further improvements in scanning speed and miniaturization, the PS-FLIm imager has the potential for clinical translational applications as a fast and compact imaging tool.
