Visible stimulated Raman scattering nanoscopy

Far-field nanoscopy of non-fluorescent metabolites in living systems remains a challenge. We introduce Ultrasensitive Reweighted Visible Stimulated Raman Scattering (URV-SRS), a label-free chemical nanoscopy technique achieving 86-nm lateral resolution and a detection sensitivity of a thousand molecules within the focal volume. We demonstrate nanoscopic imaging of natural metabolites in virus infected mammalian cells and engineered bacteria, as well as metabolic vibrational probes in the silent window.

Abstract

Far-field nanoscopy of non-fluorescent metabolites in living systems remains a challenge. We introduce Ultrasensitive Reweighted Visible Stimulated Raman Scattering (URV-SRS), a label-free chemical nanoscopy technique achieving 86-nm lateral resolution and a detection sensitivity of a thousand molecules within the focal volume. We demonstrate nanoscopic imaging of natural metabolites in virus infected mammalian cells and engineered bacteria, as well as metabolic vibrational probes in the silent window.
