Quantitative analysis of leukocyte activation in inflammatory acute conditions using digital holographic microscopy

Digital holographic microscopy (DHM) is a novel imaging technology attractive for high-throughput, label-free, and quantitative cell analysis for acute care diagnostics. This study introduces a novel DHM-based protocol to quantify leukocyte activation by examining morphological changes. Unlike traditional methods such as automated hematology analyzers, which cannot assess leukocyte activation stages, DHM enables detailed screening of leukocyte activation. By incorporating deep learning-based segmentation algorithms, DHM precisely and selectively identifies activated leukocytes, facilitating real-time monitoring of inflammatory conditions. This innovative approach aims to enhance diagnostic capabilities and support clinical decision-making at the point of care.
