Kilohertz volumetric imaging of in-vivo dynamics using squeezed light field microscopy

Volumetric functional imaging of cellular signaling and motion dynamics is challenging due to hardware bandwidth limits and restricted photon budgets in short exposure times. We introduce squeezed light field microscopy (SLIM), a computational imaging method that detects high-resolution 3D light signals rapidly using a single, low-format camera sensor. SLIM achieves over one thousand volumes per second across a 550 μm diameter and 300 μm depth field of view. Demonstrations include blood cell velocimetry in the embryonic zebrafish brain and voltage imaging of neural membrane potentials in the leech ganglion. SLIM is a robust tool for high-speed microscopy applications.
