Empowering super-resolution microscopy via engineered microscope slide for on chip patterned excitation
Structured Illumination Microscopy (SIM) is a technique developed to increase the spatial resolution in wide field fluorescence microscopy. Spatially structured light is superimposed to the spatial frequencies of the sample and a beat pattern arises. By translating and rotating the structured light over the specimen, different images can be acquired to mathematically reconstruct a super resolved one. SIM is widely employed in biological research; however, two major drawbacks, as bulkiness and low stability of the setups are present. To overcome them here we present an engineered microscope glass slide to generate the pattern directly on the sample holder.
