In vitro evidence on photobiomodulation by blue LED light

Wound healing represents a highly intricate biological process, orchestrated by a multitude of cellular and molecular mechanisms critical for preserving tissue homeostasis and safeguarding the organism against external insults. Disruptions in this process significantly increase susceptibility to severe infections and noxious agents. Among therapeutic strategies, physical modalities such as laser and LED-based interventions have gained prominence in the management of chronic wounds. Nevertheless, the underlying mechanisms of their efficacy remain inadequately elucidated, with existing literature often lacking mechanistic depth. To address this gap, in vitro investigations were conducted employing a LED device emitting blue light within the 410 to 430nm wavelength range, delivering an optical power of 1 W. Blue light doses of 4, 21, and 41 J/cm² were applied to cultured cells. Analytical methodologies, including MicroRaman spectroscopy and patch clamp electrophysiology, were utilized to interrogate the modulation of cellular redox states and membrane ionic currents following light exposure. The experimental data demonstrate that blue light exerts photobiomodulatory effects in a dose-dependent manner, underscoring its potential utility in therapeutic applications. Future research will focus on delineating the specific ion channels and intracellular signaling pathways implicated, thereby advancing our understanding of the mechanistic role of blue light in the wound healing cascade.
