Tuberculosis is persisntent bacterial infection caused by Mycobaterium tuberculosis. The types of laboratory examination currently used in the TB program was microscopic examination, culture, sensitivity test. Sputum quality is one of the determinants of the examination. Macroscopically, good quality sputum is thick, purulent, contains more than 25 leukocyte cells per field of view, has rust green color with a volume 3,5 ml per collection, and spesific smell. The study to find the determine effect of sputum quality on GenXpert moleculer rapid test and MGIT. This research uses the Post Test Only Control Group Design method with a Cross Sectional design. Samples for this research were 60 samples who unerdent both GenXperrt and Liquid media cultue. The statistical test using Chi Square. Based on the results of research, TCM results were negative, 16 (48.48%) of sputum of bad quality were negative; 17 (51.51%); Positive good quality sputum 14 (51.85); sputum bad quality , negative results were 13 (48.24%). MGIT culture of 60 samples of good quality sputum was negative, 16 (48.48%) of bad quality sputum were negative; 17 (51.51%); Positive good quality sputum 14 (51.85); sputum quality was not good, negative results were 13 (48.24%). The chi square test results for TCM and MGIT culture obtained a value of p = 0.795 (> 0.05). It can be concluded that there are not effect of sputum quality on TCM examination and MGIT culture.
