Formulation and In-Vitro Characterization of Olmesartan Medoxomil-Loaded Polymeric Nanoparticles

Introduction: The antihypertensive drug Olmesartan Medoxomil has low oral bioavailability (28%) because it is poorly soluble in water, pre-systemic metabolism, and p-glycoprotein mediated efflux. Objective: The goal of current investigation was to design Olmesartan Medoxomil loaded polymeric nanoparticles to enhance bioavailability through oral administration by using a solvent evaporation technique. Methods: The formulation comprises ethyl cellulose, polyvinyl alcohol, and Tween 80 as polymer, co-polymer, and surfactant using different concentrations and were assessed for particle dimensions, surface charge, entrapment efficiency, drug polymer interactions, powder XRD, and in vitro study. Result: The result of the study shows particle size 92.02 nm, Poly Dispersibility Index 0.167, Zeta Potential −17.2 mV, Entrapment Efficiency 94%, and release of drug 89.21%. Conclusion: It was concluded that Olmesartan Medoxomil nanoparticles shows promising approaches as a vehicle for the oral drug delivery because of a remarkable increase in solubility and enhanced bioavailability.
