Development and Evaluation of Immediate Release Tablet of Atorvastatin Calcium by Forming Inclusion Complex
Background: Several methods have been investigated recently to improve the solubility and bioavailability. Inclusion complex approach is a potential way for enhancing solubility. Aim of study: It could be said to improve the solubility of drug atorvastatin which has poor aqueous solubility by making inclusion complex with water soluble compound Sulfobutylether-β-Cyclodextrin and to improve its bioavailability. Methods: Inclusion complexes of atorvastatin and Sulfobutylether-β Cyclodextrin were formulated using kneading method of one ratio one, one ratio two, one ratio three, one ratio four and one ratio five. The solubility studies of phase tests unveil that a linear relationship between atorvastatin solubility and Sulfobutylether-β-Cyclodextrin concentrations. The tablets were formed utilizing the direct compression process. dosage forms were tested for physical characteristics like hardness, friability, and variations in tablet weight. Result: Inclusion complex can be successfully used for preparation of directly compressed tablet. In-vitro dissolution test showed formulation F5 had maximum release in comparison to marketed formulations. So, the developed formulation showed increase in solubility and bioavailability of atorvastatin. Conclusion: Currently, formulation of atorvastatin developed by using SBE-β-CD showed increased in the bioavailability thus it can be a better alternative to the presently available marketed formulation.
