Formulation, Evaluation And Development Of A Bilayer Tablet Containing Esomeprazole And Clarithromycin
The development of a bilayer tablet containing Esomeprazole and Clarithromycin presents a novel approach to improving patient compliance and therapeutic efficacy in the treatment of Helicobacter pylori infection and related gastrointestinal conditions.Esomeprazole, a proton pump inhibitor, provides prolonged gastric acid suppression, while Clarithromycin, a macrolide antibiotic, targets bacterial infections, particularly H. pylori. This study aimed to formulate and evaluate a bilayer tablet that combines the immediate release of Clarithromycin with the sustained release of Esomeprazole, optimizing their pharmacokinetic profiles and minimizing dosing frequency. The bilayer tablet was prepared by compressing granules of Clarithromycin for immediate release and Esomeprazole for sustained release, using hydroxypropyl methylcellulose (HPMC) as a matrixing agent and microcrystalline cellulose (MCC) as a disintegrant. The formulation was optimized through a series of preformulation studies, focusing on parameters such as granule size, compression force, and excipient compatibility.Comprehensive evaluations were conducted, including hardness, friability, disintegration time, and in vitro dissolution studies. The results demonstrated that the bilayer tablet exhibited acceptable mechanical strength, with a hardness of 4.7 kg and friability within acceptable limits. In vitro release studies revealed that Clarithromycin achieved rapid release within 30 minutes, while Esomeprazole maintained a controlled release profile over 10 hours, fitting the Pappas model for drug release kinetics.
