Development of lactate sensing probe with deep trench structure for analyzing cancer cells in brain tissue
Abstract To develop a real-time monitoring probe for lactate concentration in brain tissue, we have proposed a unique probe that increases detection area by deepening its structure. This probe, fabricated in a silicon substrate, features a rigid design with a finger-like deep trench to immobilize sufficient lactate oxidase to precisely detect lactate concentration within brain tissue. This article presents a micro-level lactate probe for monitoring brain tumors. An innovative deep-trench finger-like structure was designed and successfully fabricated on a silicon substrate to ensure sufficient enzyme quantities and increase the monitoring area. The manufacturing process proposed a bi-layer resist composed of parylene and photoresist, exploring the influence of the electroplating parameters of Prussian blue on detection. Biochemical detection was completed using chips with the same structure, achieving a good linear range. The detection limit also meets the requirements for lactate detection in brain tissue, validating the feasibility of this innovative design.
