Microglia Process α‐Synuclein Fibrils and Enhance their Pathogenicity in a TREM2‐Dependent Manner (Adv. Sci. 7/2025)

Parkinson's Disease In article number 2413451, Zhentao Zhang and co‐workers show that microglia process α‐synuclein fibrils to generate novel fibrils with enhanced toxicity, promoting the onset and progression of Parkinson's disease. α‐Synuclein fibrils enter microglia through TREM2. Then they are processed by asparagine endopeptidase to produce α‐synuclein 1‐103 fibrils, which are more toxic than naive α‐synuclein fibrils. These results show that blocking the aberrant activity of microglia halts the progression of Parkinson's disease. [Image: see text]
