The Impact of Small Non-Coding RNA RsaE on Biofilm Development in Staphylococcus Aureus
Biofilms of Staphylococcus aureus can adhere to a variety of medical equipment. R-Sae, a conserved S. aureus short noncoding RNA, controls central carbon flow and other metabolic processes. Our research showed how R-Sae releases biofilm matrix S. aureus isolates. Hospital patients in Hooghly, West Bengal, India, provided 250 clinical samples from December 2021 to June 2022. Culture and biochemistry detected S. aureus <tex xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">$(\mathrm{n}=60)$</tex> in clinical samples, which Vitek2 Compact confirmed. We assessed S. aureus biofilm potential using quantitative microtiter plates. The 96-well plate approach differentiated moderate, mild, and robust biofilm-producing S. aureus. Polymerase chain reaction revealed biofilm genes Ica-ADBC and R-Sae. Finally, we used real-time quantitative PCR to measure the amount of the biofilm operon Ica-ADBC and the regulatory gene RSae in S. aureus isolates. We identified S. au reus <tex xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">$(\mathrm{n}=60)$</tex> by morphology and biochemistry from 250 clinical samples. In biofilm growth, isolates were strong (25%), moderate (67%), or weak (8.3%). All isolates expressed R-Sae and the biofilm gene Ica-ADBC. The S. aureus isolates that formed strong or moderate biofilms had higher levels of IcaA, IcaD, and R-Sae than the isolates that formed weak biofilms.
