Advances in single-cell RNA-seq for nascent transcriptome and epitranscriptome
<p indent="0mm">Single-cell RNA sequencing (scRNA-seq) technology is a crucial tool for studying rare samples and cellular heterogeneity, with broad applications in life sciences and medicine. In contrast to a general description of the whole transcriptome, a detailed analysis of specific transcriptome subcomponents, such as nascent transcriptome and epitranscriptome, yields a more precise characterization of gene expression variations during cellular stress responses, as well as the distinct patterns of epi-transcriptomic modifications in heterogeneous cells, thus offering us a deeper understanding of the molecular basis of cellular functions. In this review, we highlight the unique technical advances of single-cell transcriptome library construction in the fields of single-cell nascent transcriptome sequencing and single-cell epitranscriptome sequencing, and discuss the future development trends and prospects.
