Transdermal Delivery of Resveratrol by a Nanomicelle-Loaded Enzyme-Triggered Hydrogel for Diabetic Wound Healing

Wound infections can lead to serious diabetes complications that are difficult to cure. Despite the great efforts made, exploration of bioactive patches to effectively promote diabetic wound healing remains a challenge. Here, we propose an enzyme-triggered self-assembly oligopeptide hydrogel-embedded cell-penetrating peptide modified on the surface of a resveratrol-loaded nanomicelle (Res-CNM/Gel) to enhance the delivery of resveratrol for wound healing. The porous structure and composition of peptide-based hydrogels allow them to have great drug loading capacity and biocompatibility. Due to the utilization of an enzymatic self-assembly gel formation strategy, the hydrogel can be transformed at 37 °C to prolong drug release. In addition, the resveratrol-loaded nanomicelle conferred the ability of the hydrogel to inhibit inflammation and promote cell migration. Results showed that Res-CNM/Gel prolonged drug release actively and promoted wound healing by reducing inflammation, promoting cell migration, and inhibiting bacteria. Therefore, Res-CNM/Gel possesses great potential for diabetic wound healing in the future.
