A new approach to PDT dosimetry using an image-guided surgery affibody probe
Photodynamic therapy (PDT) can be used to assist image guided surgical resection for head and neck cancers. Here, we use the image guided surgery probe ABY-029, an EGFR-targeted fluorescent affibody protein conjugated to IRDye800CW, as a dosimeter for PDT of 3D tumor nodules using TLD1433. Using MOC1 3D nodules as an in vitro model that simulates microscopic residual disease, we showed a strong inverse relationship between ABY-029 fluorescence intensity bound to the MOC1 3D nodules and the TLD1433-PDT dose applied to the nodules. This approach has the potential to guide the dosimetry of intraoperative PDT following image guided surgery.
