Analysis of artocarpin content and hepatoprotective activity of ethanol extract of mobe (Artocarpus lacucha Buch-Ham.) leaves on the liver of rats induced by paracetamol
Liver dysfunction due to the use of hepatotoxic drugs such as paracetamol is still a problem, especially in the long term and in high doses. Natural products derived from plants are often used as hepatoprotective agents because of their potent antioxidant properties. Our study reports that ethanol extract of Mobe leaves ( Artocarpus lacucha Buch-Ham) has the potential as a hepatoprotective agent. The artocarpin content that we analyzed using HPLC in Mobe leaves allows this activity. The test was carried out on paracetamol-induced rats by observing the decrease in ALT, AST, ALP, total bilirubin, total protein, LDH, and an increase in SOD levels in the blood serum of rats. The image of liver hepatocytes and the expression of IL 6, Caspase – 3, and TNF-α genes were also observed. The results showed that the administration of ethanol extract of Mobe leaves had lower levels of ALT, AST, ALP, total bilirubin and higher total protein compared to the group given 0.5 % Na-CMC suspension induced by paracetamol. Increased SOD levels and decreased LDH also occurred, followed by histopathological observations of the liver showing normal liver conditions and no inflammation even though induced by paracetamol. The percentage of Caspase – 3, IL-6, and TNF-α also experienced significant differences with the 0.5 % Na-CMC group. The content of secondary metabolites in Mobe has excellent potential in protecting and improving liver function. These findings are expected to be used as a guide to explore the benefits of the Mobe plant as a new protective agent.
