Ganoderma lucidum polysaccharides relieve High Glucose-induced human renal tubular epithelial cell damage

Objective Ganoderma lucidum polysaccharides (GLP) have biological and pharmacological actions in mammals, but its mechanism on high glucose (HG)-triggered human renal tubular epithelial cell (RTEC) injury is still unclear. Methods HK-2 cells were divided into Control, HG, GLP-I (HG + 3mg/L GLP), GLP-II (HG+ 6mg/L GLP), GLP-III (HG + 12mg/L GLP), GLP-II+PMA group. Cell proliferation and apoptosis were monitored using CCK-8 and flow cytometry. MDA, SOD, and ROS levels were determined using kits. Western blot assessed protein expression. Results Cell viability, Bcl-2, and ROS level in HG-treated HK-2 cells were enhanced by GLP exposure, whereas apoptosis rate, Bax and p65 expression, and MDA and ROS level were reduced. PMA treatment might partly abolish the effects of GLP on HG-induced cell damage. Conclusion GLP prevents HG-caused human RTEC damage by inhibiting NF-κB signal.

Abstract

Objective Ganoderma lucidum polysaccharides (GLP) have biological and pharmacological actions in mammals, but its mechanism on high glucose (HG)-triggered human renal tubular epithelial cell (RTEC) injury is still unclear. Methods HK-2 cells were divided into Control, HG, GLP-I (HG + 3mg/L GLP), GLP-II (HG+ 6mg/L GLP), GLP-III (HG + 12mg/L GLP), GLP-II+PMA group. Cell proliferation and apoptosis were monitored using CCK-8 and flow cytometry. MDA, SOD, and ROS levels were determined using kits. Western blot assessed protein expression. Results Cell viability, Bcl-2, and ROS level in HG-treated HK-2 cells were enhanced by GLP exposure, whereas apoptosis rate, Bax and p65 expression, and MDA and ROS level were reduced. PMA treatment might partly abolish the effects of GLP on HG-induced cell damage. Conclusion GLP prevents HG-caused human RTEC damage by inhibiting NF-κB signal.
