Revalidation of external validity of red blood cells indices by population-based data

Objective: The objective of current study was to revalidate the external-validity of the red-blood-cell indices in the population. Study Design: cross-sectional study. Place and Duration: Collection and testing of blood samples have been done in Husaini Hematology Blood Bank and Basic Medical Sciences Laboratory of Faculty of Pharmacy, Hamdard University, Karachi. from 15thJanuary to 30thApril 2023. The study includes 8,134 blood samples (N). Methodology: After taken ethical approval of study and informed consent; collected blood samples were analyzed by Hematology Analyzer, Sysmex XP-100 and XN–1000. Main outcome measures were levels of hemoglobin, hematocrit, microcytosis, macrocytosis. 22nd version of Statistical Package for Social Science was used for analysis of data. Results: Statistically significant differences were noted for reference-ranges of female to their below-normal-ranges of hemoglobin (p=0.001), hematocrit (p=0.032), MCV (p=0.011), MCHC (p=0.014), MCH (p=0.021), RBC-count (p=0.0111); above-normal-ranges of hemoglobin (p=0.01), hematocrit (p=0.0001), MCV (p=0.001), MCHC (p=0.003), MCH (p=0.01), RBC count (p=0.033). Significance also noted in references-ranges for male to their below-normal-ranges of hemoglobin (p=0.021), hematocrit (p=0.039), MCV (p=0.019), MCHC (p=0.022), MCH (p=0.038), RBC count (p=0.010); above-normal-ranges of hemoglobin (p=0.0145), hematocrit (p=0.0001), MCV (p=0.016), MCHC (p=0.029), MCH (p=0.0001), RBC-count (p=0.001). Conclusion: Reference ranges for RBC indices are re-validated of a JCI (Joint Commission International) accredited and state of the art tertiary care hospital and have high external validity.
