16S rRNA sequence for identification bacteria recorded a high rate of hemolysis
Aims:The study determines the rate of hemolysis activity against human, sheep and goat blood by supernatant of different clinical isolates by micro-titration methods.16S rRNA is then used to identify the highest hemolysis-producing isolates and conduct relationships between them by a phylogenetic tree.Methodology and results: Respiratory infections associated with respiratory devices constitute one of the most dangerous and essential problems at the hospital acquired infections level, so this has become a starting point for the current study.Eighty clinical isolates were obtained from various Respiratory devices. The ability of hemolysin production by microorganisms was performed depending on the 96 micro-titer plate method using various kinds of blood (human, sheep and goat). The results of this study show that Pseudomonas aeruginosa is the most common species and the highest hemolytic species among other isolates, as 33 isolates of P. aeruginosa were obtained from 80 isolates with 100% hemolysis activity against blood.Sequence analysis technique of 16S rRNA had been accomplished on four isolates of Pseudomonas spp. with the highest value of hemolytic activity against humans, sheep and goats and recorded three of it in NCBI with accession number as ON799272.1,ON799399.1,ON872231.1 withdrawing the evolutionary tree to determine the relationships between it.Conclusion, significance and impact of study: Pseudomonas aeruginosa was the best isolate in the hemolysis of blood and it was found that goat's blood was the best type for hemolysis with the highest rate.
