Research on the Mechanism of Zexie Decoction in the Intervention of Obesity Base on Network Pharmacology and Molecular Docking Technology
To use network pharmacology and molecular docking technology in predicting the main active ingredients and targets of Zexie Decoction treatment in obesity and explore the potential mechanisms of its multi-component-multi-target-multi-pathway. Methods: the active components and corresponding targets of Zexie Decoction were obtained through tcmsp platform, the obesity related targets were obtained with the help of genecards and OM IM database, and the intersection of drug targets and disease tar gets was obtained. We used Cytoscape software to construct the "active component target" network diagram, used string platform to draw PPI network, used mcode plug-in for protein cluster analysis, used metascape database for go enrichment analysis and K EGG pathway analysis, and used autodock and PyMOL for molecular docking. 515 targets of Zexie decoction, 1307 targets of obesity and 166targets of drug and disease were obtained. Through go analysis, Through go analysis, 425 biological process items, 129 molecular function items and 56 cell component items were obtained; through KEGG pathway analysis, Serotonergic synapse, Insulin resistance and TNF signaling pathway were obtained. The results of molecular docking showed that the main active components of Zexie Decoction in the treatment of obesity were Alisol A, Alisol B, Biatractylolide and Alisol A 24-acetate , which had goo d binding with core proteins AKT1, MAPK3,SRC,MAPK1,PTGS 2 and EGFR. This study preliminarily revealed the potential mechanism of Zexie decoction in treating obesity with multi-component, multi-target and multi-channel, which laid a foundation for further study on the mechanism of Zexie decoctionin treating obesity. Zexie decoction has the effect of multi-component, multi-target and multi-channel treatment of obesity.
