Network Pharmacology of Trichopus Zeylanicus ssp. Travancoricus Burkill Ex K. Narayanan Leaf Extract Revealed CA2, ERN1, TTR, and SNCA as Inducible Genes

Indigenous herbaceous medicinal plant Trichopus zeylanicus ssp. travancoricus Burkill ex K. Narayanan, present in the Agasthyamalai hills in India, is considered a health rejuvenator best known for their pharmacological properties. Despite its well-known potential, no comprehensive reports exist regarding the bioactive-induced genes and their associated physiochemical properties. In this study, we focused on identifying potent genes (biomarkers) from the plant bio-actives and their physiochemical properties. Inducible targets were identified from the Binding DB, while gene ontology studies and associated pathways were determined through the TOPPGENE and KEGG web servers. Network enrichment analysis was performed by using Cytoscape 3.3.0. Phytoconstituent’s (PCs) physicochemical properties were ascertained through ADMETlab 2.0. Among the total of 22 PCs, four specific PCs - 1,2-Benzenediol, Vanillin, Isovanillic acid, and Gamma-hydroxy isoeugenol were linked to predicted targets. They are CA2, CA4, CA6, ERN1, TTR, CA14, SNCA, PF3D7_0932300 and yrdA. Notably, SNCA, ERN1, CA2, and TTR were determined as the most prominently induced genes (biomarkers). Furthermore, none of the PCs violated the physicochemical properties criteria. The PCs 1,2-Benzenediol, Vanillin, Isovanillic acid, and Gamma-hydroxy isoeugenol hold potential for medicinal applications and offer prospects for the development of effective drugs targeting conditions associated with alpha-synuclein (SNCA), Serine/threonine-protein kinase/endoribonuclease IRE3 (ERN1), Carbonic anhydrase 2 (CA2) and Transthyretin (TTR).
