Protective Effects of <i>Cuscutae Semen</i> Extract from Oxidative Stress and Amyloid <i>β</i>25-35-Induced SH-SY5Y Neuronal Cell Dysfunction
Objectives: Amyloid beta (A) is generated by the amyloidogenic pathway. Its accumulation is a major cause of Alzheimer's disease. The present study investigates the free radical scavenging activities and inhibition of A 25-35 -induced neurotoxicity by Cuscutae semen (CS) water extract treatment. Methods: The scavenging activities of radicals, including 2, 2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl (OH), and nitric oxide (NO) of the CS water extract were assessed. Moreover, the SH-SY5Y cells were used to investigate cell viability, inflammation-related protein expression, and inflammatory cytokine secretion using CS water extract to determine neuroprotective effects. Results: The CS water extract inhibited the production of free radicals such as DPPH, OH, and NO, dose-dependently.Meanwhile, oxidative stress was induced, and cell viability was decreased by A 25-35 .The CS water extract treatment significantly increased cell viability in A 25-35 -stimulated SH-SY5Y cells.Additionally, the treatment with CS water extract significantly down-regulated the expression of inducible nitric oxide synthase and cyclooxygenase-2 in A 25-35 -stimulated SH-SY5Y cells.Secretion of interleukin-1 and interleukin-6 in A 25-35 -induced SH-SY5Y cells was significantly inhibited by CS water extract. Conclusions: This study indicated the free radical scavenging activities and neuroprotective effects of CS water extract.
