Characterization of RNASE L, Ankyrin Repeat Domain mutation Gene of RNASE L Gene in Gastric Cancer patients in Pakistani Population
Gastric carcinoma is the fourth most common type of cancer and the second leading cause of cancer death in the world. And mortality rate are still high with more than 1 000 000 cases in2018. . According to Worldwide data 2018, Around one in 12 of all oncological deaths are caused by gastric cancer. It has the5th ( fifth) highest occurrence amongst cancers, 5.7% of all young people infected with the disease. Gastric cancer is too individual of the behaviorally influenced, and therefore preventable of the cancers Gastric cancer has highest risks which have been reported in the central Africa, Eastern Asia and Western Africa. Gastric cancer patients 80% to 90% that has been observed which initiation of the cancer is due to the H pylori. Many types of the gene mutations are too involved in Gastric cancer pathogenesis. RNASE L is the important significant roles in the antiviral response. That is controlled by interferon and the involved in cleavage of viral RNA for that reason the aim of this research study is to identify RNASE and ANK domain mutation/s of RNAS E L gene in gastric cancer patients. Methodology Total 120 blood samples was collected in this study. DNA extracted through the PCR. To analyze .the RANSE-L amplified PCR product the gel-electrophoresis performed. After that The amplified samples of RANSE-L was sent to the directed Macrogen, Korea for the sequencing although analysis of mutation done through Bio Edit and MEGA11 software.. All mutations of RNASE-L gene associated with H pylori . Result The types of our research sample were; 1: GC patients positive, 2: Patients of GC with H-pylori positive and 3: diagnosed patients of H-pylori . Out of 120 samples of blood, 5%, of GC, 50% are of GC alongside with H pylori, and the residual samples of blood are of H-pylori positive individuals. Of these, 58.3% are men and 41.6% are women. Sequence examination revealed that one (1) only sample had SNV(single nucleotides variant) G>A in the domain of RNASE, and not of any one sample had SNV in domain ARD . Conclusion In this research study, we tried to analyze genetically feature of GC therefore which it can support us therapeutically. Our healthcare organization is in very necessity of certain immediate improvements which are essential to control peoples of GC
