Exploring The Potential Of Colloidal Lipid Carrier For The Delivery Of Curcumin As Polyphenolic Nutraceuticals Useful In Cancer Chemoprevention

This project work is focused to engineer a colloidal lipid carrier to improve the solubility, bioavailability, bioaccessibility of the said nutraceuticals for the management of cancer of curcumin, a poorly water-soluble drug. Curcumin0-NLC were prepared by using high pressure homogenisation technique. Three factor-two level (23) factorial design was found useful to identify the variables influencing the responses. 3 operating variables i.e. an lipid (mg, X1), power (X2, W) and the time (X3, min) while Z-average (size, Y1), PDI (Y2) and the ZP (mV, Y3). The optimized batch (batch C7) showed spherical morphology and had Z-Avg (57.13± 2.3nm), PDI (0.194) and ZP of -26.5mV. PXRD confirmed a partial amorphization of drug and lipid with change in crystalline order of lipid matrix during NLC processing. The Curcumin loaded NLC was found to follow Peppas release kinetics. In cytotoxicity study against RAW-264.7 macrophages, lipid, and oil disclosed certain cytotoxicity against RAW-264.7. However the presence of curcumin in NLC suppresses toxicity. In this regard the influence of the surface behavior of stabilizers and significant role of curcumin in inflammatory immune response was thought significant.
