Acute Dermal Toxicity Study of “Doorvadi Cream” by using Wistar Albino Rats according to OECD-402

In Ashtangsangrah second chapter specially dedicated to treatment of child ailments explained one disease under the name of Paridagdhchavi which means in this condition look of childlike burn like. For this condition advised one doorvadi lepa. Female Wistar albino rats were used in an in vivo investigation at the Toxicology Department, Institute for Industrial Research & Toxicology (IIRT), Ghaziabad. The objective of the study was to assess DC (Doorvadi Cream) acute dermal toxicity (ADT) study by following OECD test no 402. The starting dose of 2000mg/kg body weight was selected for range finding study. The test sample was directly applied on clipped skin of a single rat and observed immediately after dosing at least once during the first 30 minutes, periodically during the first 24 hours, with special attention given during the first 2 to 6 hours after the beginning of the exposure period and daily thereafter, for a total of 14 days. As no evident toxicity was found, the main test was conducted at dose level of 2000mg/kg body weight. The animals were closely observed daily for clinical signs of toxicity and mortality for a period of 14 days. The body weight was recorded on day 0 (pretreatment), 7th and 14th (post treatment). The necropsy was performed on all animals. Results obtained from present investigation concluded that the test sample “Doorvadi Cream” was found non-toxic with dermal application upto the dose of 2000mg/kg body weight. Hence the test sample was classified as Category IV (LD50 >2000 mg/kg body weight)
