DNA barcoding of Pelawan Kepuh (Tristaniopsis obovata Benn) utilizing the rbcL Gene

Abstract This research proposed the first DNA barcoding of Tristaniopsis obovata Benn. collected from Bangka Belitung through the analysis of the rbcL gene. The DNA sequences have been submitted to the GenBank, establishing a primary reference for Pelawan Kepuh ( Tristaniopsis obovata Benn) from Bangka Island, based on rbcL region. Sample collection was carried out in Pelawan Forest, within the Biodiversity Park in Namang, Bangka Tengah Regency. The DNA extraction, PCR amplification, and DNA sequencing were performed as molecular analysis. Total 483 bp sequence length were obtained and has a 99.4% similarity with Tristaniopsis sp. according to BLASTN analysis. Furthermore, evolutionary history analysis exhibited that Pelawan Kepuh, is grouped with other species within the Tristaniopsis genus. The rbcL barcode analysis highlights its less effectiveness in species identification within the Tristaniopsis genus at both genetic and broader taxonomic levels. Further studies were essential to identify Tristaniopsis obovata Benn using other DNA barcoding regions, including ITS for a comprehensive identification of Pelawan Kepuh.
