Effect of Ethanol Extract of Okra Fruit on NF-ΚB Expression in Wound Healing Wistar Rats With Diabetes Mellitus
Objectives:The main objective of the study was to see the effect of Okra Fruit Ethanol Extract on NF-κB Expression in the Healing of Wistar Rat Wounds in the Diabetes Mellitus Model. Theoretical Framework: NF-κB is a critical component in wound healing due to its role in regulating inflammatory responses, cell proliferation, angiogenesis, and tissue remodeling. However, regulation of its activity must be balanced, as excessive activation can slow the healing process or cause complications, especially in conditions such as diabetes mellitus. Therefore, therapeutic approaches targeting NF-κB are a promising area of research in the management of chronic wounds. Method: Experimental laboratory research , type of research Randomized Control Trial (RCT) and post test only control design approach . . Wistar rats were made into DM with a sample size of 30 consisting of, negative control (K-), positive control (K+), Experimental group with a dose of P1 dose of 300 mg / KgBB, P2 dose of 600 mg / KgBB and P3 dose of 900 mg / KgBB okra fruit ethanol extract. The rats were wounded in the back area with a size of 1 cmx 1 cm. On the 21st day, blood was taken through the retroorbital vein to examine NF-kB levels. –in Center Between University (PAU) Gajah Mada University, using the enzyme linked immunosorbent assay (ELISA) method. Analyzed using the One Way Analysis of Variance (Anova) parametric test. Results and Discussion : The results of the study showed significant differences between groups. The negative control group (537.56) was significantly different from groups P1, P2 and P3, all showing a decrease in Nf-kB expression. Okra dose of 300 mg/KgBB (123.57) showed a significant decrease in NF-kB expression compared to the negative control, but higher than the doses of 600 mg/KgBB and 900 mg/KgBB. Okra dose of 900 mg/KgBB had significantly lower NF-kB expression compared to the okra group dose of 300mg/KgBB. Ethanol extract of okra fruit showed a significant reduction effect on NF-kB expression in wounds. Research implication: This study provides information that okra fruit ethanol extract has the potential to reduce NF-kB expression, which functions in the inflammatory process in diabetes mellitus wounds. Originality / value :The results of this study indicate that ethanol extract of okra fruit is able to reduce the expression of inflammatory biomarkers, namely NF-kB. As well as a new perspective on the use of traditional plants as wound healing therapy.
