P26 Analysis of the detectability of the association of polymorphism of genotypes of the GSTP1 gene in patients with psoriasis

Abstract The search for genetic factors in the development of chronic dermatoses is a priority in medical science. It should be noted that in the development and pathogenesis of psoriasis, along with the main trigger factors, so-called modifier genes are involved, the effect of which is largely determined by environmental factors. Among these genes, of particular interest are the glutathione-S-transferase genes—GST, which are responsible for the biotransformation of chemical, biological agents, drugs, etc. entering the body. Particular attention is paid to the enzyme of the second phase of detoxification, glutathione transferase, a product of the GSTP1 gene. The purpose of our research was to assess the detectability of the association of polymorphism of genotypes of the gene for the second phase detoxification enzyme GSTP1 in patients with psoriasis. Material and research methods Forty-five patients with psoriasis aged 18–49 years were examined. Among them, there were 18 male and 27 female patients. All patients underwent clinical, molecular genetic and statistical studies. The control group consisted of 64 healthy individuals of the corresponding age. Research results Analysis of molecular genetic studies of the frequency distribution of the association of polymorphism of genotypes IIe 105 Val of the xenobiotic enzyme GSTP1 gene showed an increased frequency of detection of the heterozygous variant—in 44.4% (20/45) of cases, while in the control group of healthy individuals, it was 10.9% (7/64) respectively. [χ2 = 21.78, P < 0.0003; odds ratio (OR) = 6.51; 95% confidence interval (CI) 2.44–17.375]. The mutant homozygous variant of the IIe 105 Val polymorphism of the GSTP1 gene was detected in 6.6% of cases (3/45), respectively (χ2 = 21.78, P < 0.0003; OR = 10.62; 95% CI 0.54–210.93). Conclusion The results of molecular genetic studies showed that allelic variant G and hetero/homozygous genotypes of the IIe 105 Val polymorphism of the GSTP1 gene are significant markers of an increased risk of developing severe forms of psoriasis in Uzbekistan (P < 0.05) (χ2 = 22.56, P < 0.0002; OR = 7.02; 95% CI 2.89–17.06). The functionally favourable genotype A/A of the GSTP1 gene is a reliable protective marker against the development of pathology (χ2 = 21.78, P < 0.0003; OR = 0.12; 95% CI 0.04–0.31).
