10001- ANGI-1 TRIAL OF MULTI-OMICS ANALYSES USING PAIRED SAMPLES FROM NEWLY DIAGNOSED AND RECURRENT GLIOBLASTOMAS DURING BEVACIZUMAB THERAPY

Abstract BACKGROUND We explored multi-omics analyses using surgical samples obtained from patients received preoperative neoadjuvant bevacizumab therapy (neoBev) for newly diagnosed glioblastoma (nGBM) and its recurrence (rGBM). MATERIALS AND METHODS Of all eight surgical samples from two cases treated by neoBev and two cases without preoperative Bev therapy (naiveBev) at nGBM and rGBM were analysed. RESULTS Compared with naiveBev, expression levels of VEGF, HIF-1 alpha, FOXM1 in neoBev were downregulated, whereas CD163 was upregulated. By comparison between long and short overall survival (OS), Foxp3/CD8 ratio and FOXM1 were decreased in long OS. The transcriptional factor induced under hypoxia was significantly elevated in naiveBev with long OS. DISCUSSIONS AND CONCLUSIONS Analyses by space multi-omics provided novel findings correlated with specific gene expressions and anatomical location focused on tumor microenvironment, which might be quite useful to comprehend the mechanism of effectiveness and resistance of targeted therapies including antiangiogenic or immunotherapies. But problems were listed as follows; cumbersome interpretation of huge amount of information for which might be essential of co-operation by skillful bioinformaticians, and huge amount of cost. Those problems especially for cost-effectiveness should be solved for the development of this field of clinical and basic research.
