516 Intrinsic factors regulating hair growth: insights into growth factors, cytokines, chemokines, and interferons

the Republic of) Senescent cells can accumulate within tissues and induce the senescence of surrounding normal cells through the senescence-associated secretory phenotype (SASP), thereby accelerating organ aging.Senolytics are drugs that can selectively eliminate these senescent cells, whereas senomorphics are another class of drugs that modulate senescence phenomena within tissues.Unlike senolytics, senomorphics do not directly kill senescent cells.Instead, they aim to delay or block the onset of senescence. The senolytic effects of red ginsengderived components (RGCs) on senescent skin cells remain unknown. Therefore, we sought to determine whether RGCs could be a candidate for senolytics in aging skin by assessing if RGCs have an anti-aging effect through senolysis on senescent primary human keratinocytes and dermal fibroblasts.Senescent cell models were created using repeated ultraviolet B irradiations and a UV senescent cell-normal cell co-culture system to determine the impact of SASP from senescent cells on neighboring normal cells. Each of three RGC (the total extract, saponin, and non-saponin) was treated to determine whether senolysis was induced and to evaluate the changes in senescence markers (p16), SASP factors (IL-6 and IL-8), epidermal differentiation, and collagen synthesis. As a result, there were no significant senolytic effects on RGCs.However, the senescence marker (p16) and SASP (IL-6, IL-8) as well as the inflammatory factor (IL-1b) were attenuated after treatment with the RGCs.This indicates an improvement in cellular aging and a reduced risk of spreading inflammatory aging to surrounding normal cells.Additionally, the differentiation of epidermal cells and the collagen synthetic ability of fibroblasts were also improved to some extent. In summary, while RGCs may not function as a senolytic, it has been verified to mitigate skin cell aging and enhance cellular function through anti-aging mechanisms distinct from senolysis.Notably, Red Ginseng-derived Components exhibit potent senomorphic properties, making them effective agents for preventing skin aging.
