Decline in forced vital capacity (FVC) in subjects with idiopathic pulmonary fibrosis (IPF) and progressive pulmonary fibrosis (PPF) compared with healthy references

Introduction: IPF and PPF are associated with decline in FVC. Aim: Compare baseline FVC and decline in FVC in subjects with IPF in the INPULSIS trials and with PPF in the INBUILD trial with values from hypothetical healthy references. Methods: Baseline FVC and changes in FVC at week 52 were assessed in the nintedanib and placebo groups of the pooled INPULSIS trials and the INBUILD trial. Values were compared to those in hypothetical healthy references matched for age, sex, race and height, derived using equations published by the ERS Global Lung Function Initiative. Results: In the INPULSIS trials, mean (SD) baseline FVC (mL) was 2714 (757) in the nintedanib group vs 3597 (725) in their healthy references and 2728 (810) in the placebo group vs 3624 (703) in their healthy references. In the INBUILD trial, mean (SD) baseline FVC (mL) was 2340 (740) in the nintedanib group vs 3411 (786) in their healthy references and 2321 (728) in the placebo group vs 3354 (807) in their healthy references. Over 52 weeks, FVC decline was about 7-fold greater than in their healthy references in the placebo groups, and only 3 to 4-fold greater than in their healthy references in the nintedanib groups (Figure). Conclusions: These data show the physiological impact of IPF and PPF and support the clinical relevance of the reduction in FVC decline seen with nintedanib. <fig><object-id>erj;64/suppl_68/PA673/F1</object-id><object-id>F1</object-id><object-id>F1</object-id><graphic></graphic></fig>
