Development of a method for the isolation and purification of anti-IFN-gamma interferon autoantibodies and quantitative detection
Backgroud: Anti-γ interferon autoantibody (AIGA) syndrome serves as one of the newly discovered anti-cytokine autoantibody diseases. characterized by a large production of neutralizing AIGA production and disseminated infection by opportunistic pathogens in Asian populations. Methods: The sensitivity and specificity of chromatography-based clinical diagnostic methods for AIGAs were evaluated by a clinical cohort using Genomic-based single-cell sequencing, tandem affinity chromatography to obtain polyclonal AIGAs, and BLI technology for affinity evaluation. Result: Isolated extracts by tandem affinity chromatography-based methods possessing neutralizing activity against IFN-γ with an average affinity of 3.374E-10 and complete competitive binding activity. The cut-off value for the chromatography-based AIGAs assay was initially determined from a clinical cohort: 0.12063 mg/ml, with 96.7% sensitivity and 96.6% specificity. Conclusion: A novel absolute quantitative method for the diagnosis and continuous monitoring of AIGAs was developed and achieved good sensitivity and specificity.
