Initiation of dupilumab led to reduced use of oral corticosteroids (OCS) and other medications over 12 months in patients with chronic rhinosinusitis with nasal polyps (CRSwNP): A US real-world practice study

Background: Evidence on dupilumab effectiveness in patients with CRSwNP in US clinical practice is limited. Aims and Objectives: To compare OCS and other medications use in CRSwNP patients before and after initiation of dupilumab in the US. Methods: A retrospective observational cohort study in adults with CRSwNP who initiated dupilumab 300 mg every 2 weeks between June 2019 and June 2022. Data from the OM1 Real-World Data Cloud and the Reg-ENTSM Registry were used. In this analysis, CRSwNP-related OCS use (within 5 days of a CRS and/or NP diagnosis or within 30 days of sinus surgery) and bursts (OCS use for ≤14 days with ≥7 days gap between bursts), and prescription of intranasal corticosteroids (INCS) and antibiotics in the 12 months before/after dupilumab initiation (pre-/post-dupilumab periods, respectively) are summarized descriptively. Results: The overall cohort comprised 1016 patients. The proportion of patients with CRSwNP-related OCS use decreased from 59.1% pre-dupilumab to 17.7% post-dupilumab; 316 (31.1%) patients had OCS bursts (mean [SD] 1.5 [0.9] per patient) pre-dupilumab vs 143 (14.1%; 1.2 [0.5]) post-dupilumab; 76.5% of patients with CRSwNP-related OCS use pre-dupilumab had no CRSwNP-related OCS use post-dupilumab. INCS use decreased from 50.5% to 31.1% and antibiotic use from 64.6% to 31.8%. Conclusions: Patients with CRSwNP who initiated dupilumab had reduced OCS and other medications use during the 12 months following dupilumab initiation compared with the 12 months pre-dupilumab.
