Sirtuins as biomarkers of accellerated cellular senescence in patients with Idiopathic Pulmonary Fibrosis (SIR-IPF)

Background: Idiopathic Pulmonary Fibrosis (IPF) is a progressive lung disease characterized by deposition of fibrotic tissue in the lungs. Sirtuins a family of proteins involved in different enzymatic reactions might have a role as biomarker of accellerated alveolar type-II senescence. Methods: We present a case-control study, involving IPF patients followed at ILD Clinic U.O.C. Vanvitelli, Monaldi Hospital, Naples, Italy. Results: 34 IPF patients and 19 age and gender-matched controls were recruited. Study population characteristics are reported in Table 1. Sirt-1 was lower in IPF versus controls (p <.001) while Sirt-3 was higher in IPF versus controls (p<.001). Sirt-1 was positively correlated with FEV1, FVC and DLco(Fig. 1); no significant association was present for Sirt-3. IPF patients with more severe GAP Stages expressed lower levels of Sirt-1 compared with GAP Stage I. Conclusions: This study provides novel data about the role of Sirtuins as biomarkers of accellerated senescene in IPF. <table-wrap><object-id>erj;64/suppl_68/PA3367/TB1</object-id><object-id>T1</object-id><object-id>TB1</object-id><table><colgroup><col></col><col></col><col></col><col></col></colgroup><tbody><tr><td></td><td>IPF</td><td>Controls</td><td>p-value</td></tr><tr><td>Age</td><td>75.0[69-77.1]</td><td>71.0 [67-77.0]</td><td>0.218</td></tr><tr><td>Gender</td><td>26(76.3)</td><td>12(63.2)</td><td>0.302</td></tr><tr><td>Antibrotics</td><td>30(88.2)</td><td>.</td><td>.</td></tr><tr><td>GAP Index</td><td>4 [3-5]</td><td>.</td><td>.</td></tr><tr><td>FEV1%</td><td>89.0 [77.0-102]</td><td>.</td><td>.</td></tr><tr><td>FVC%</td><td>81.0 [64.0-92.1]</td><td>.</td><td>.</td></tr><tr><td>DLCO%</td><td>46.9 [32.0-61.3]</td><td>.</td><td>.</td></tr><tr><td>6MWD</td><td>252 [208-396]</td><td>.</td><td>.</td></tr><tr><td>Sirt-1</td><td>667[435-858]</td><td>925[794-1173]</td><td><.001</td></tr><tr><td>Sirt-3</td><td>338[230-500]</td><td>154[99.8-246]</td><td><.001</td></tr><tr><td>Sirt-6</td><td>242[146-301]</td><td>263[238-328]</td><td>0.131</td></tr><tr><td>Sirt-7</td><td>4742[3622-6302]</td><td>4957[2112-5519]</td><td>0.522</td></tr></tbody></table></table-wrap> <fig><object-id>erj;64/suppl_68/PA3367/F1</object-id><object-id>F1</object-id><object-id>F1</object-id><graphic></graphic></fig>
