Real-world NGS testing results and treatment modalities in lung cancer: Data update from LALUCA - Landsteiner Lung Cancer Research Platform

Introduction Lung cancer is a major health burden in Austria. However, there is limited real-world data on patient characteristic and NGS testing results. The LALUCA registry gathers high-quality data in a clinical setting and thereby provides a tool for real-world investigations of lung cancer. Aims and objectives Here, we aim to describe real-world NGS testing results and treatment modalities in lung cancer patients within the LALUCA registry. Methods Lung cancer patients diagnosed between 11/2020 and 12/2023 were included in this analysis. The focus of the present analysis lies on histological and molecular pathological diagnostics and treatment modalities. Results The prevalence of histological subtypes (n=1022) was as follows: adenocarcinoma (54.6%), squamous cell carcinoma (20.7%), small cell lung cancer (SCLC) (13.6%), non-small-cell lung cancer-not otherwise specified (NSCLC-NOS) (9%), and other neuroendocrine tumors (0.9%). Molecular pathological diagnostics (n=788) showed that the most common mutations were KRAS (27.7%) and EGFR (11.3%). With a prevalence up to 3.8% mutations or fusions including ALK, BRAF, HER2, MET exon 14 skipping, NTRK, RET and ROS were observed. Patients treated with curative treatment intent (n=225) received neo-adjuvant therapy in 72.4% and adjuvant therapy in 24.9%. Palliative treatment (n=520) consisted of chemo-immuno-therapy in 46.7%, followed by targeted therapy in 19.6%, immunotherapy in 16.5% and chemotherapy in 16.9% alone. Conclusions The LALUCA registry offers an important platform to investigate real-world histological and molecular pathological diagnostics and treatment modalities in lung cancer.
