Late Breaking Abstract - Clinical efficacy of Tezepelumab in pre-selected non-Type 2 asthma patients
Introduction Stromal Lymphopoietin (TLSP) plays a role in the non-Type 2 (T2) asthma pathway, which makes this biomarker a target for the treatment. Blockade of TSLP reduces a broad spectrum of biomarkers and cytokines associated with airway inflammation. Tezepelumab is a monoclonal antibody (IgG2λ) directed against TSLP and prevents interaction with the heterodimeric TSLP receptor. Tezepelumab has a probable positive effect on exacerbation rate, lung function, and quality of life in patients with non-eosinophilic asthma. This is the first case series that highlights the clinical effect of tezepelumab in pre-well-defined non-T2 asthma patients following the Global Initiative for Asthma (GINA) guideline. Method Patients were treated (per protocol) with 210 mg of Tezepelumab (subcutaneous) every 4 weeks for 6 months. During each visit (T0-T6), the patients received a lung function (FEV1) and 2 questionnaires; the Asthma Quality of Life Questionnaire (ACQ) and the Asthma Quality of Life Questionnaire (AQLQ). Medical ethical approval for this study was obtained (NL78940.100.21). Results All patients were females and aged between 18 to 68 years (Table 1). The BMI ranged from 19.90 to 39.80 kg/mm2. After treatment with Tezepelumab, the FEV1 improved for 6 out of the 8 patients and the ACQ- and AQLQ-score improved for 5 and 6 out of the 8 patients, respectively (Figure 1). Conclusion This is the first case series demonstrating that patients with non-T2 asthma benefit from treatment with Tezepelumab regarding lung function, ACQ, and AQLQ. However, an extensive placebo-controlled trial is needed to confirm the clinically beneficial effects of Tezepelumab in non-T2 asthma patients.
