Nemolizumab is a mAb that inhibits the signaling of the neuroimmune cytokine IL-31, which promotes inflammation and pruritus in atopic dermatitis. Silverberg et al (Lancet 2024;404(10451):445-60; https://doi.org/10.1016/S0140-6736(24)01203-0) conducted 2 randomized, placebo-controlled phase 3 clinical trials (ARCADIA 1 and 2) to assess the safety and efficacy of nemolizumab in adult and adolescent patients with moderate-to-severe atopic dermatitis. Nemolizumab improved skin clearance, sleep disturbance, and pruritus after 16 weeks of treatment as compared to placebo in both trials.
