Bioremediation of Kalamkari Dye using Novel Bacterium Bacillus Inaquosorum Isolated from Textile Contaminated Waste Soil of Srikalahasti, A. P
Dye contamination by textile industries is imminent hazardous that severely impact environment has great global attention. Due to diverse nature of textile dyes that consist of variety of pollutants like dyes, acids, bases, metal salts. Microorganism' s remediation of these dyes are often quite challenging using eco friendly method. The purpose of present study was undertaken the use of bacteria isolate and identified from contaminated site of kalamkari industry of Srikalahasti, Andhra Pradesh, India. Isolation bacterial strains using dilution method, and 13 different isolates were screened. Morphological Characterization of Isolated Bacteria isolates. Identification of Isolated Bacteria by 16S r RNA molecular sequencing . growth parameter optimization to enhance further dye degradation with different concentration 125, 250, 500, 750 and 1000 mg/l, pH ranging from 5 to 9 and temperature varies from 20 to 500C for 48 hrs at max. Wavelength 325nm. Hence, in the present study, thirteen bacterial strains were isolated from kalamkari effluent contaminated site of Srikalahasti, screened for dye degradation and bacterial isolate-1 showing significant activity with 57.07± 0.34%. Bacterial isolate-1 selected for optimization indicated optimum pH of 5.0, temperature of 45 ° C and dye concentration of 250mg/l. Dye degradation of dye was enhanced from 57 to 81% after optimization. Identification of bacterial isolate-1 by 16S rRNA gene sequencing showed 93% sequence similarity with Bacillus inaquosorum (NRRL B-23052). Bacterial isolates habituating the effluents of textile industries play important role in bioremediation of dye which can be further enhanced by optimization of their growth parameters.
