Transferability of SSR Markers Developed from Gooseberry to Black and Red Currants

Eleven microsatellite markers previously developed from gooseberry sequences were tested on red and black currants. The results showed amplification of all microsatellite loci on black currant accessions, while on red currant accessions, there was no amplification at three loci (RucANS, RucDFR2-1, RucDFR1-3). Polymorphic loci were identified in both black and red currants. In the MTT-7 locus of the studied black currant genotypes, amplification of three fragments was observed, suggesting duplication of this locus. At the same time, in the MTT-7 locus of red currant, amplification typical of a monolocus microsatellite was observed. The RucHLH-1 and RucUFGT loci were tested on a hybrid red currant family (Belaya Potapenko × 1426-21-80). Using linkage mapping approach, the RucHLH-1 locus was mapped to the linkage group 4 of the red currant genome, and the RucUFGT locus (presumed), to the linkage group 1. The set of microsatellite loci for the genus Ribes is currently limited. This study shows that some of the SSR markers developed from gooseberry are amplified and reveal polymorphism on currants too, and can also be used to study both black and red currants.
